Here's a good reference: About the screens you see when your Mac starts up. What you are seeing is the Prohibitory symbol. The drive you have in your system doesn't have a working system.
This could be caused by selecting the wrong drive selected or, your drive/SATA cable is failing. Try starting your system up with Boot manager so you can make sure you've selected the correct drive, then try diagnostic mode and run Disk Utility see if the drive is working.
