iMac Intel 21.5 inch-EMC-2308
Imac started to boot up with the chime and the display would flash briefly and then go to black. For a while I was able to put the iMac in target mode and run repair permissions from another laptop, but I can't do this anymore. I tried a new power supply and that did not solve the problem.

The flashing and going black sounds like an LED Backlight Board issue to me. I'd hook it up to an external monitor, boot from the original system installation disk, run disk utilities.

Thanks! I cannot get the external monitor to show anything but black. I am not sure if I am using the correct key sequence to enable the external monitor or do I need one?
If your computer is generating a video signal you don't need to do anything to activate an external monitor. No external signal would indicate a video card failure. In the upper right corner of this link shows the part number you may want: http://www.welovemacs.com/mb950lla.html
